My mother raised me when I was a kid growing up, so that would make her the number one woman in my life. There is nothing another woman can do to surpass that.

My mother passed on years ago, but she will always hold a spot in my heart. My wife is here today and influences my happiness more than any other person on earth and NO SHE IS NOT REPLACEABLE!

I think it does depend on the situation... If the day was special for one of them, or if their need was greater, than that woman becomes #1. For example, on Mother's Day, mom is #1 while my girlfriend (I'm not married yet) is #1 on her birthday. But if there was a point of contention between the two, then my GF / wife would be who I stand behind (unless their stance is completely opposite to my values...)

In a life and death situation, I'd choose my mother over my girlfriend easily.  However, I'd side with my girlfriend in the majority of her disputes with my mother.
